Capture 5023 cells for control and 10107 cells for FGF2 organoids using the 10x Chromium System Libraries were prepared according to 10x Genomics Chrmoium Next GEM Single Cell 3' GEM Kit instructions (Cat# 1000130). Briefly, single cells were emulsified in 10X Chromium system and each cell's RNA was barcoded. cDNA was amplified, fragmented and indexed primers 10-13 cycles and library fragments of ~400 bp. The cDNA was flowed onto a Illumina flow cell for cluster generation. Libraries were sequenced on 4 lanes of a Illumina NextSeq500 for high output PE75 v2.5 following the manufacturer's protocols. scRNA-Seq / Drop-Seq
