Treatment protocol |
BALB/c mice were percutaneously infected with 30±2 cercariae of S. japonicum. Successful infection was confirmed by detecting serum specific antibody or identifying parasitic eggs in fecal samples after infection.

Mice were sacrificed under anesthesia and spleens were harvested. Spleen tissue was minced in a sterile Petri plate. The erythrocytes were lysed in ACK buffer and a single-cell suspension of murine splenocytes was prepared by passing cells through 70 μm strainers. Single cell RNA libraries were prepared using Chromium Single cell 5' Library & gel bead kit and coupled Chromium single cell V(D)J enrichment kit&scT/BCR library according to the manufacturer's protocol
