Treatment protocol |
HUVECs were cultured in complete medium. For normal glucose, 25 mM mannitol was added. For high glucose, cells were treated with 25 mM glucose and 5 ng/ml TNFa. Cells were harvested after 3 and 7 days.

Cells were fixed with 1% formaldehyde for 10 minutes. After quenching and cell lysis, chromatin was sonicated using a Bioruptor (Diagenode). The libraries were constructed using the KAPA HyperPrep kit following the user manual.
