Extraction protocol |
RNA was extracted and quality control was performed as mentioned above.We performed RTqPCR, using progeny from 3 wells of 96 well plates for RNA extraction using the GenElute Mammalian Total RNA Miniprep Kit (Sigma-Aldrich) according to the manufacturer’s instructions with addition of DNAseI from the On-Column DNase I Digestion kit (Sigma-Aldrich). Library preparation was performed with the LexoGen Quant Seq. Kit. Libraries were sequenced on an Illumina HiSeq4000 sequencing system.

Data processing |
Single-end reads were generated, with an average of 4 million reads obtained per sample. Alignment was performed with Hisat2. Raw counts were log2 transformed before performing downstream analyses.
