Title |
PBS treated nasal mucosa of wild type mouse (C56BL/6) |
Sample type |
RNA |
Source name |
nasal mucosa of wild type mouse
Organism |
Mus musculus |
Characteristics |
genotype/variation: wild-type strain: C56BL/6 treatment: PBS
Treatment protocol |
Flagellin and other inhibitors were administered intranasally. For intranasal (i.n.) instillations, 25 ul of solution were applied drop-wise into each nostril.
Growth protocol |
Mouse nasal mucosa were harvested from 8wk mouse
Extracted molecule |
total RNA |
Extraction protocol |
nasal septum and turbinate’s mucosa from each side was gently elevated off the cartilage and bone and total RNA was isolated from these tissues using TRIzol
Label |
biotin
Label protocol |
Samples were enzymatically fragmented and biotinylated using the WT Terminal Labeling Kit (Affymetrix)
Hybridization protocol |
Fragmented end-labeled cDNA was hybridized to the GeneChip® Mouse Gene 1.0 ST arrays for 16 hours at 45 “C and 60 rpm as described in the Gene Chip Whole Transcript (WT) Sense Target Labeling Assay Manual (Affymetrix)
Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G
Description |
PBS treated nasal mucosa of wild type mouse (C56BL/6)
Data processing |
Data was normalized using the RMA(Robust Multi-Average)algorithm implemented in Affymetrix expression console version 1.1 software probe group file: MoGene-1_0-st-v1.r4.pgf meta-probeset file: MoGene-1_0-st-v1.r4.mps
