Extraction protocol |
Total RNA was isolated at 12-24 hours after editing using RNeasy Micro Kit (QIAGEN), and DNase treatment was performed using RNase-free DNase Set (QIAGEN), according to the manufacturer’s instructions. RNA was quantified with The Qubit 2.0 Fluorometer (Thermo Fisher) and its quality was assessed by a 2100 Agilent Bioanalyser (Agilent Technologies). Minimum quality was defined as RNA integrity number (RIN) of >8. 20-100 ng of total RNA were used for library preparation and subsequent ultra-low input or standard sequencing by Genewiz (Azenta Life Sciences).
