Growth protocol Bone marrow cells from C57/BL6 mice were cultured with GM-CSF and IL-4 for 6 days and stimulated with LPS for 24 hours to generate bone-marrow derived DC (BMDC). CD11c+ BMDC were isolated by anti-mouse CD11c magbeads to generate maDC. maDC were then cocultured with spleen stroma cells for 9 days to generate diffDC. And then diffDCs were transfected with si-Suclg2 or si-nc.

Extraction protocol diffDCs transfected with si-Suclg2 or si-nc were collected for RNA extraction, total RNA was extracted with TRIzol reagent.
cDNA library construction and sequencing were performed by Beijing Genomics Institute using Illumina HiSeq platform.
