Growth protocol |
Bone marrow cells from C57/BL6 mice were cultured with GM-CSF and IL-4 for 6 days and stimulated with LPS for 24 hours to generate bone-marrow derived DC (BMDC). CD11c+ BMDC were isolated by anti-mouse CD11c magbeads to generate maDC. maDC were then cocultured with spleen stroma cells for 9 days to generate diffDC. And then diffDCs were transfected with si-Suclg2 or si-nc.

Extraction protocol |
diffDCs transfected with si-Suclg2 or si-nc were collected for RNA extraction, total RNA was extracted with TRIzol reagent. cDNA library construction and sequencing were performed by Beijing Genomics Institute using Illumina HiSeq platform.

Data processing |
cDNA library construction and sequencing were performed by Beijing Genomics Institute using Illumina HiSeq platform. High-quality reads were aligned to the mouse reference genome GRCm38.p4 (mm10). The expression levels for each of the genes were normalized to fragments per kilobase of exon model per million mapped reads (FPKM).
