Treatment protocol The R-HL60 cell line was established through the continuous treatment of parental HL60 cells with increasing concentrations of cytarabine.
Growth protocol Cells are maintained in RPMI 1640 medium supplemented with 10% fetal bovine serum and 1% antibiotic–antimycotic reagents under a humidified atmosphere of 5% CO2 at 37 °C.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from HL60 and R-HL60 cells using TRIzol Reagent (Thermo Fisher Scientific, Waltham, MA, USA) and according to the instruction manual. The RNA samples were submitted to Welgene Biotech. Co., Ltd. (Taipei, Taiwan, R.O.C) for gene expression array analysis. RNA quantity was determined using an ND-1000 spectrophotometer (Nanodrop Technology, Wilmington, DE, USA) and RNA quality was verified using an Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA).
Label Cy3
Label protocol Total RNA (200 ng) was amplified using a Low Input Quick Amp Labeling Kit (Agilent Technologies, Santa Clara, CA, USA) and labeled using fluorescent Cy3 dye (Agilent Technologies, Santa Clara, CA, USA). After incubation with fragmentation buffer at 60°C for 30 min, 0.6 µg Cy3-labeled cRNA was fragmented to an average size of 50–100 nucleotides.
 
Hybridization protocol Correspondingly fragmented and labeled cRNA was pooled and hybridized to a SurePrint G3 Human Gene Expression 8x60K Microarray (Agilent Technologies) at 65 ◦C for 17 h.
Scan protocol The Cy3 microarray was scanned at 535 nm with a microarray scanner (Agilent Technologies, Santa Clara, CA, USA).
Data processing The scans were analyzed using Feature Extraction 10.7.3.1 software (Agilent Technologies, Santa Clara, CA, USA).
The data were normalized by quantile normalization.
The normalized_data.txt file includes normalized signal intensities.
