Treatment protocol |
We transplanted Balb/c left lung to C57BL/6iPHIL recipient with co-stimulatory blockade, then administrate either Diphtheria Toxin or Saline since 30days post-engraftment. Harvest the lung allografts after 3-4 weeks eosinophil depletion period.

Lung tissue was minced and digested by IMDM medium containing collagenase II and DNAse for 35 minutes at 37°C in a shaker. The digested lung tissue was passed through a 70-μm cell strainer and treated with ACK lysing buffer to remove red cell contamination. Library was performed according to the manufacter’s instructions (Single Cell 3' v3, 10X Genomics)
