Bone marrow cells were harvested from STAT5KO and WT control mice, single ESLAM HSCs was FACS sorted into 2.3ul 0.2% Triton-100 containg RNAse inhibitor Tagmentation is carried out using the Illumina Nextera XT DNA sample preparation kit. The protocol is based on the Tagmentation protocol from Fluidigm.

RNA-seq reads were aligned to mm10 + ERCC92 using GSNAP (version 2018-03-25) with parameters (-B 5 -t 24 -n 1 -Q -N 1). Reads in features were counted with htseq-count (HTSeq version 0.9.1) with the parameter (-s no). Assembly: GRCm38.81 Supplementary files format and content: HTSeq counts text file with raw counts and htseq metrics Supplementary files format and content: Anndata h5 file with filtered cells and genes and data transformations (in layers)
