Total RNA was prepared from cells or cell derived extracellular vesicles using miRNeasy Serum/Plasma advanced kit (Qiagen).

For each experiment, 100 ng RNAs were reverse treanscribed and followed by generation of cDNAs. The resulting cDNAs were transcribed and labled with cyanine 3-CTP and the labeled antisense cRNAs were purified using RNAeasy columns (QIAGEN).

Hybridization protocol |
The purified antisense cRNAs were hybridized to Agilent’s 8 × 60,000 microarrays at 45 °C for 16 hrs.

Scan protocol |
Hybridized microarray slides were washed and scanned on the Agilent DNA Microarray Scanner (G2505C) using one color scan setting for 8x60k array slides

Data processing |
The scanned images were analyzed with Feature Extraction Software (Agilent) to obtain background subtracted and spatially detrended Processed Signal intensities. Features flagged in Feature Extraction as Feature Non-uniform outliers were excluded.
