Treatment protocol Cells derived from respective cultures maintained with specific glucose concentrationswere trypsinized, passaged and cultured supplemented with either 5 mM or 25 mM glucose for 24 h followed by increasing the concentration of glucose supplementation to 50 mM in both sets of cultures (5 → 50 and 25 → 50) for the next 24 h afterward. Briefly, an additional amount of D-Glucose was added to supplement for the additional 45 mM glucose or the additional 25 mM glucose in the two culture systems in parallel, to represent a hyperglycemic-induced stress condition consisting of 50 mM glucose in total, for 24 h.
Growth protocol The human cardiomyocyte (CM) cell line AC16 was maintained in DMEM containing 10% fetal bovine serum , 100 U/mL penicillin and 100 mg/mL streptomycin and cultured with either 5 mM or 25 mM D-glucose in a humidified incubator at 37°C with 5% CO2 for at least 3-passages prior to them being used for experiments.

Data processing Sequence cluster identification, quality prefiltering, base calling and uncertainty assessment were done in real time using Illumina's NCS 1.0.2 and RFV 1.0.2 software with default parameter settings. Sequencer .cbcl basecall files were demultiplexed and formatted into .fastq files
Sequenced reads were trimmed for adaptor sequence and aligned by STAR aligner
Reads per kilobase of exon per million reads mapped (RPKM) were calculated.
