Mettl3KO or Pulsed control cells were resuspended in Trizol and total RNA was extracted with Direct-Zol RNA mini Prep (Zymo Research)
RNA libraries for RNA-seq were prepared using Tecan Revelo mRNA Library Prep Kit following manufacturer's protocols.

| Data processing |
Quality control of reads was perfromed using fastqc v.0.11.9 and RSeQC v.4.0.0 and mapped to reference mouse genome build Mus_musculus.GRCm39.107 using HiSat2 v.2.2.1.
Gene expression counts were generated using featureCounts v.2.0.1 and corrected for batch effect using the removeBatchEffect function in R.
Differenatial expression analysis was performed using the Omics Playground platform.

Supplementary files format and content: *.gene.counts.txt files contain raq read counts for each samples.
