All experimantal samples were grown in growth factor-free NSPC medium (with addition of 10% FBS), for 24 hours, in CO2 incubator at 37C

For cell growth and propagation: NSPCs were grown in serum-free NSPC medium and ECs were grown in EC medium (Celprogen), at 37C, in CO2 incubator

After 24 hours in transwell co-culture, the ECs and NSPCs were separated and total RNA was isolated using RNeasy Mini Kit (Qiagen)

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100ng total RNA (Affymetrix 3' IVT Express labeling kit).

Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Mouse Genome 430 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G

Data processing |
The data were analyzed with RMA and Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
