RNA |
Source name |
Diphtheria-Toxin treated+ 24h recovery
Organism |
Mus musculus |
Characteristics |
genotype: Atp4b-DTR tissue: Gastric Corpus Tissue
Treatment protocol |
Diphtheria toxin (225ng/mouse) or saline vehicle was administered via ip injuection daily for three days. Mice were euthanized at 24, 36, 48, or 72 hours after final siphtheria toxin injection
Growth protocol |
Mice were grown in a temperature and light controlled mouse facility; littermate controls were used when possible. Adult mice age 8-12 weeks were used.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA extracted, purified by RNEasy Micro Kit, and RNA intergrity verified on Agilent Bioanalyzer
Label |
Biotin
Label protocol |
Total RNA was extracted and used to prepare biotinylated cDNA with Affymetrix WT plus reagent kit
Hybridization protocol |
Labelled cDNA hybridized to Affymetrix GeneChip Mouse Gene 2.0 ST Arrays according to manufacturers instructions and placed in GeneChip Hybridization Oven 620 for 18hr at 45C. Microarrays were washes and stained with Affymettrix Fluidics Station 450.
Scan protocol |
Arrays were scanned with a GeneChip® 7G Scanner 3000 laser confocal slide scanner
Data processing |
Microarray data data expression was process using Command Console (Affymetrix, Inc) and the raw .CEL files were analyzed using Partek Genomic Suite v7.0 (Partek Inc., St Louis, MO). Microarray CEL files were imported into Partek and normalized using RMA.
