Treatment protocol Growth factor supplements were replaced as of 50% in every second day and passaging was performed every 4/5th day. In culture, ENP-derived neurospheres were passaged every 5th days into a single cell suspension (P = passage).
Growth protocol ESSCs were cultured in 25 cm2 flasks at a density of 1 X 105 to 3 X 105/ ml in neural basal medium (Stem cell technologies) with the previously discussed growth factor conditions and incubated at 370c, 5% CO2.
Extracted molecule total RNA
Extraction protocol RNA samples were isolated from triplicates of three groups (treatment A, D & E), using Qiagen miRNeasy Mini Kit.
Label Biotin
Label protocol 1000 ng of total RNA was Poly (A) tailed and ligated to a biotinylated signal molecule using flash tag biotin HSA RNA Labeling Kit.
Hybridization protocol The biotinylated target RNA sample was then hybridized to Affymetrix miRNA arrays for 16 hours at 480C with rotation at 60 rpm according to GeneChip Hybridization. The GeneChip® miRNA Array probeset was used for miRNA profiling.
Scan protocol The scanned images were inspected for hybridization efficiency and, CEL files generated from AGCC (Affymetrix GeneChip Command Console) were imported into miRNA quality control QC Tool software for determination of data quality.
Description miRNA expression data from Group E-derived ESSCs
Group E ESSCs
Data processing miRNA array data analysis was performed with the Partek Genomics SuiteTM software.
For evaluation of miRNA expression data, Affymetrix CEL files were imported into the software prior to analysis.
Expression values calculated for rno-miRNAs.
