The total RNA was extracted from the silk gland using TRIzol (Invitrogen, Carlsbad, CA, USA). 1 µg of total RNA was used for the construction of sequencing libaries. The RNA libraries were validated according to the manufacturer’s instructions.

The original data were analyzed using fastp version 0.19.5, and clean data with Q30 > 92.93% was obtained from each sample Compare clean data(reads) with reference genome. Use assembly software StringTie Version 1.3.3b for reference-based assembly. The differentially expressed genes were determined at |log2 (fold change)| > 1 and p-adjust < 0.05 using DESeq2 Version 1.24.0 Assembly: StringTie Supplementary files format and content: tab-delimited text file includes raw counts for each Sample Supplementary files format and content: tab-delimited text files include RPKM values for each Sample
