Treatment protocol |
T-ALL xenografted mice were treated for 6h with either 40µg control IgG2a, or OKT3 or Teplizumab. Mice were killed and leukemic cells were flow cytometry-sorted (hCD45+; hCD7+).

Growth protocol |
NSG female mice were tail vein injected (1 million cells/mouse) with human T-ALL cells (hCD45+; hCD7+) and treated when tumor burden reached around 1% in blood.

Single strand cDNA targets were prepared starting from 200 ng of total RNA (Rneasy kit QIAGEN) using the Ambion WT Expression Kit according to Affymetrix recommendations.

Single-stranded cDNA were labeled with the Affymetrix GeneChip WT Terminal Labeling Kit according to Affymetrix recommendations.

Hybridization protocol |
3 μg of cDNAs were hybridized for 16 hours at 45C on GeneChip Human Gene 2.0 ST arrays (Affymetrix).

Scan protocol |
The chips were stained in the GeneChip Fluidics Station 450 (Affymetrix) and scanned with the GeneChip Scanner 3000 7G (Affymetrix) at a resolution of 0.7 µm.

Data processing |
Raw data were extracted from the scanned images using the Affymetrix GeneChip® Command Console (AGCC) version 4.1.2. CEL files were further processed with Affymetrix Expression Console software to calculate probe set signal intensities using Robust Multi-array Average (RMA) algorithms with default settings. RMA expression estimated from expression Console software version 1.4.1. RMA algorithms with default settings.
