The J558-allografted BALB/c female mice were orally administered 0.5% methylcellulose as vehicle or TAK-931 at 60 mg/kg, QD for 6 days.

| Growth protocol |
J558 cells were maintained in DMEM supplemented with 10% Horse Serum and penicillin/Streptomycin in humidified atmosphere with 5% CO2 at 37C. The female BALB/c mice were inoculated subcutaneously with 5 × 10e5 J558 cells each (in 0.1 mL, no Matrigel) in the right flank.

The J558 allografted tumor samples were sliced into small pieces (2–4 mm3) and then isolated into single cells using Tumor Dissociation kit (Miltenyi Biotec). The cells were filtlated with 70 mm and 30 mm niron mesh. After centrifugation, the supernatant was discarded, suspended in 1xRBC lysis Solution (Invitrogen), centrifuged with 10% FBS/DMEM, and suspended in 10% FBS/PBS.
Library was performed according to the manufacter's instructions (Single Cell 3’ GEM v3.1, 10x Genomics).
