Extraction protocol The fully growing oocytes (FGO) were harvested from WT and Gdf9-cre Nat10 conditional knockout female mice 36-48h after PMSG injection, then after washing by PBS with 0.2% containing, the oocytes were lysed with lysis buffer containing 0.2% Triton X-100 and RRI.
The library construction were standard Smart-seq2 protocol. Briefly, polyA RNA in cell lysis were first hybrided with Oligo-dT primer, then reverse transcripted by the PrimeScript Ⅱ RTase and TSO primer. The producted cDNA were pre-amplified with KAPA HiFi HotStart Ready Mix (2×) & TSO-PCR primer. Purified intact cDNA were then fragmented and amplified with barcodes adding by using TruePrep DNA Lybrary Prep Kit V2 for Illumina (cat.#TD502, Vazyme).

Data processing The raw reads were first cleaned with Trim Galore v0.6.7, and the cleaned reads were further mapped to mm10 genome by using STAR v2.7.10a. Uniquely mapped reads were applied for gene expression quantification by using FeatureCounts b2.0.2. The trimmed reads were also mapped to the ERCC genome for gaining the percentage of ERCC.
