Status Public on Jul 10, 2024
Title iRGCs, Ctrl_c
Sample type SRA

Source name RGC-like neurons (iRGCs)
Organism Mus musculus
Characteristics cell type: RGC-like neurons (iRGCs)
genotype: control
Treatment protocol MEFs were infected with Ascl1+Brn3b+Islet1 overexpressing lentiviruses, doxycycline was added on day 0 to induce exogenous gene expression, cells were switched to neuronal medium (DMEM/F12+GlutaMAX+N2+B27) on day 2 of reprogramming; medium was half-changed every other day.
Growth protocol MEFs were cultured in DMEM+10% FBS, at 37℃ in incubator.
Extracted molecule polyA RNA
Extraction protocol RNAs were extracted using TRIzol reagent
NEBNext UltraTM RNA library Prep Kit for Illumina following manufacturer's protocol

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing Basecalls performed using CASAVA
clean reads were aligned to the mouse reference genome (mm10) using Hisat2
featureCounts was used to count the reads numbers mapped to each genes
Assembly: mm10
Supplementary files format and content: read counts for each genes were given in the Excel file
