Growth protocol Y. lipolytica transformed with the whole-genome sgRNA plasmid library was inoculated in SD-Leu medium. When cell growth reached the late exponential phase (after approximately 36 hours), 1 mL of culture was collected for plasmid extraction.

Extraction protocol Plasmid extraction was performed using ZymoprepTM Yeast Plasmid Miniprep II (Zymo Research Corporation).
Sequences containing spacers in sgRNA plasmids were amplified via PCR, purified, fragmented, and their ends were repaired and reformed to become clusters.

Data processing The original data of sequencing results were subjected to image Base calling by using the software CASAVA(v1.8.2), and the quality was preliminarily analyzed.
The overlap regions of paired-end sequencing results were merged.
Through sequences alignment, the abundance of spacer of every sgRNA plasmid could be determined.
