Growth protocol |
Auricula auricula-judae is cultured in PDA at 25°C for 14 days, exposed to gamma radiation or not, and then inoculated into a sawdust medium mixed with sawdust and rice bran in an 8:2 (w/w) ratio. After growing the mycelium at 25°C for 30 days, the fruit body is grown by adjusting the temperature to 18°C and humidity to 90%.

RNA was extracted from the homogenized fruit bodies using the Ribospin II RNA purification kit (GeneAll Biotechnology, Seoul, Korea). 1,000 ng of total RNA was used of the construction of sequencing libraries. cDNA was synthesized using the cDNA synthesis kit (BIOFACT, RT PRE-MIX, Daejeon, Korea).
