Description

CNTN6 is a neural adhesion molecule of the contactin subgroup of the immunoglobulin superfamily.


Cloning and Expression

By probing a cerebellum cDNA library with a rat Nb3 fragment, Kamei et al. (1998) isolated a cDNA encoding CNTN6, which they called NB3. The deduced 1,028-amino acid GPI-anchored membrane protein is 43% and 44% identical to F3 (CNTN1; 600016) and TAG1 (CNTN2; 190197), respectively, and 90% identical to the rat protein. It contains hydrophobic segments in the N and C termini representing, respectively, a potential signal peptide and a GPI-anchoring signal. The N terminus also possesses 6 repeats typical of a C2-type Ig-like domain, while the C terminus has 4 repeated motifs homologous to type III fibronectin repeats. Northern blot analysis revealed ubiquitous expression in brain regions of a 3.7-kb transcript, with highest levels in cerebellum, followed by thalamus and subthalamic nucleus, and lower levels in corpus callosum, caudate nucleus, and spinal cord.

Lee et al. (2000) characterized the developmental expression pattern, promoter region, and regulation of Cntn6 in mouse brain.


Mapping

Using FISH, Kamei et al. (1998) mapped the CNTN6 gene to chromosome 3p26-p25, near the CNTN3 gene (601325).
