choice-of-anchor M domain-containing protein

This model represents a domain about 180 amino acids long, often repeated and sometimes cyclically permuted, containing a signature region described by TIGR03769 (which is just 41 residues long). Many members of this family have either LPXTG or PEP-CTERM domains at the C-terminus, for covalent anchoring at the cell surface by sortases or exosortases, respectively. Other member proteins that lack such C-terminal regions may instead have lipoprotein signal peptides at the N-terminus. The domain is duplicated in members of TIGR03773, such as CmrA2. The related protein TroA (see TIGR03772), with regions homologous to most of the length of this model, appears to have a function related to zinc uptake.

TIGR03769 domain-containing protein

This HMM a bacterial surface protein that is often duplicated. The domain was first observed occurring one to three times in Actinobacterial proteins, some of which have LPXTG-type sortase recognition motifs for covalent attachment to the Gram-positive cell wall. The region is now recognized as a "choice-of-anchor" domain, since multiple members, covering a broad taxonomic range, can be found anchored by sortases (e.g. LPXTG proteins) or exosortases (e.g. PEP-CTERM proteins).

anchored repeat ABC transporter, substrate-binding protein

Members of this protein family are ABC transporter permease subunits as identified by PF00950, but additionally contain the Actinobacterial insert domain described by TIGR03769. Some homologs (lacking the insert) have been described as transporters of manganese or of chelated iron. Members of this family, such as the zinc-regulated protein TroA ( transport related operon A) of Corynebacterium diphtheriae, typically are found along with an ATP-binding cassette protein, a permease, and an LPXTG-anchored protein with two or three copies of the TIGR03769 insert that occurs just once in this protein family.
