This family contains integral membrane proteins that are related to Drosophila rhomboid protein Swiss:P20350. Members of this family are found in bacteria and eukaryotes. Rhomboid promotes the cleavage of the membrane-anchored TGF-alpha-like growth factor Spitz, allowing it to activate the Drosophila EGF receptor. Analysis has shown that Rhomboid-1 is an intramembrane serine protease [2][3][4] (EC:3.4.21.105). YqgP, as found in Bacillus subtilis, is a rhomboid family intramembrane serine protease with an N-terminal cytosolic domain that can bind Mn2+ and Zn2+, and a C-terminal TPR repeat region. YqgP acts directly as a protease to cleave the magnesium transporter MgtE when high zinc or manganese concentrations risk toxicity. But YqgP also acts as a "pseudoprotease", an adaptor for the membrane-bound ATP-dependent processive metalloprotease FtsH, which further degrades MgtE, among its many substrates.
