Design: Brain Heart Infusion broth was inoculated with a single colony of E. coli and incubated for 4 to 6 h at 35C. Genomic DNA was isolated from 400 L of culture using the Maxwell 16 Cell SEV DNA Purification kit (Promega, Madison, WI). DNA was then quantified using the Quant-iT High-Sensitivity DNA Assay Kit (Life Technologies Inc., Burlington, ON). Sequencing libraries were constructed from 1 ng of gDNA using Nextera XT DNA Sample Preparation and Index Kits (Illumina, Inc., San Diego, CA) according to manufacturers instructions. Genomic sequencing was performed on the Illumina MiSeq Platform (Illumina, Inc.) using a 600-cycle MiSeq Reagent kit v3 (Illumina, Inc.).
