Design: After whole genome amplification samples were qualified, randomly interrupted using a Covaris ultrasonic disruptor, and then the entire library preparation is completed through the steps of end repair, A-tailing, sequencing adapters, purification, and PCR amplification. After the library is constructed, Qubit 2.0 was used for preliminary quantification, library was diluted, and then we use Agilent 2100 to detect the inserts in the library. After the insert size meets expectations, use the Q-PCR method to accurately quantify the effective concentration of the library. After the library is qualified, different libraries are pooled to flowcell according to the effective concentration and target data volume requirements, the cBOT is clustered and sequenced using the Illumina high-throughput sequencing platform.

After recovering the species whole genome, different types of molecular markers such as the mitochondrial genome, single-copy nuclear gene, and ultra-conservative components will be extracted. The phylogeny of the superfamily will be reconstructed and the relationship between major groups will be revised and clarified, making possible to explore the evolution patterns of important morphological features.
