mRNA sequencing (mRNA-seq) was conducted in the three closely-related Drosophila species, i.e., D. miranda, D. pseudoobscura, and D. obscura to compare the transcriptomes and gene expression level.

Construction protocol: Total RNA was extracted from 5 female 3rd instar larvae of D. miranda after removing imaginal discs but containing genital imaginal disc. mRNA was then purified by using oligo dT beads. Strand specific library was constructed with NEB #E7420. Sexing was based on the visual identification under microscopy and further confirmed by PCR with Y chromosome specific primers after the extraction of genomic DNA from same individuals.
