show Abstracthide AbstractA high resolution linkage map of goldfish(Carassius auratus) was constructed based on transcriptome. The samples contained 79 F2 offsprings and their parents, their transcripts were sequenced using Illuminia Hiseq2000 platform.For each individual in 79 progenies and two parents, 12 organs including scale, skin, muscle, eye, brain, liver, kidney, spleen, heart, blood, intestine and gonad were collected for total RNA extraction. Total RNA was extracted from each tissue separately using Trizol reagent (Invitrogen, USA), and then treated with RNase-free DNaseI (NEB, USA) to remove genomic DNA contamination. For each individual, equal quantities of total RNA from each of the twelve tissues were pooled. The pooled total RNA sample was analyzed on a Bioanalyzer 2100 (Agilent Technologies, Santa Clara, CA, USA) for RNA integrity. 3µg of pooled total RNA from each individual were used for RNA-seq library construction.
