Design: The high-quality RNA samples were used for both mRNA/lncRNA and small RNA library preparation using MGIEasy RNA Library Prep Set. In brief, for mRNA/lncRNA library preparation, the following steps were carried out: (i) rRNA depletion was performed to enrich mRNA/lncRNAs, (ii) reverse transcription, end repair, 3-end addition of adenine (A), adaptor ligation to RNA fragments and purifications were carried out according to kit instructions, (iii) rolling circle replication amplification, single strand separation and cyclization steps were performed then, (iv) DNA nanoball synthesis and sequencing were performed on MGISEQ-2000 (MGI, Shenzhen, China) platform at pair-end 100 bp reads.

In this project, B. terrestris was used to determine how expression of chemosensory genes vary depending on the basis of four developmental stages; larvae, early-stage-pupa, late-stage-pupae and adult by using RNA-seq. miRNAs and lncRNAs, which play a key role in regulating the expression of these genes, were also investigated. These results will enhance our understanding of the sense of smell in insects and will guide us to realize new techniques and practices for agricultural pollination.
