Design: The wild type (WT) Zhonghua16 and its spontaneousseed coat crack and brown testa color mutantĄŽpscbĄŻwere planted in the same field (Wuhan, China). Seed coat samples were taken at 20, 40, and 60 days after flowering (DAF) from ten different seedlings in 2014. Twelve representative seeds were sampled from each seedling at each developmental stage of both the wild type and the mutant. Three biological replications were designed. The testa separated from the sample seeds was sliced. The sliced WT testa samples and the pscb mutant samples were frozen rapidly in liquid nitrogen and kept at -80Ąć. A portion of the samples was used for RNA extracting using the Tiangen RNA extraction kit (category number). A portion of the sample of DAF60 was used for quantification of lignin, anthocyanin, proanthocyanidin and phytomelanin.
