Design: RNA was extracted from samples from four colonies and subsequently pooled (310 mites for the V. destructor cDNA libraries) using TRI reagent (Sigma-Aldrich) according to the manufacturer’s instructions in a Geno/grinder homogenizer (Metuchen, NJ, USA). The extracted RNA was further purified by precipitation with 2.5 M Lithium Chloride. The samples were washed in 70% ethanol, dissolved in RNase-free sterile water and stored at −80 °C until it was used. The quality and quality of the extracted RNA was evaluated using an Agilent bio-analyzer.
