Design: We extracted mRNA from the testes of 5 small male H. rufescens, synthesized strand-specific cDNA, fragmented it, ligated adapters and amplified, then sequenced with 76-bp paired-end reads on an Illumina Genome Analyzer.

Abalone is a long-studied model to study rapid evolution in fertilization. Our goal was to discover new proteins present in sperm that may be important for the evolution of sperm-egg interaction. Testis mRNA was sequenced via strand-specific cDNA in order to study the testis transcriptome of Haliotis rufescens. This transcriptome was used as a database to search mass spectra from shotgun proteomics of H. rufescens sperm.
