Design: Total RNA extracted using Trizol (Invitrogen) according to manufacturer's instructions, ribosomal RNA removed using the Ribo-Zero rRNA Removal Kit (Human/Mouse/Rate; Illumina) according to manufacturer's instructions, and strand-specific RNA-Seq libraries synthesized using the NEBNext Ultra Directional RNA Library Prep kit (NEB), with the optional User Enzyme step to selectively degrade the 2nd strand before PCR amplification. NB: this sample was spiked in with 1/10th mass of C. elegans RNA (N2 strain) before library synthesis
