Design: Total RNA was isolated from the female brain of different adult Asian arowana individuals (Tong Yan hybrid) using TRIzol according to the manufacturer’s protocol. RNA integrity and size distribution were checked with Bioanalyzer 2100 (Agilent technologies, Santa Clara, CA, USA). Concentrations of all the female brain samples were standardized to 200ng/mL, and equal volumes of female brain total RNA from different individuals were combined into one pool for library construction and high throughput sequencing using a 454 GS FLX Titanium platform by Macrogen (Seoul, Korea).
