Design: 1. Biological material Sitophilus oryzae weevils are reared on wheat grains at 27.5 C and at 70% relative humidity. The Bouriz strain was because it is free of any facultative symbionts, including Wolbachia, and it harbors only S. pierantonius. Septic injuries were made on fourth instar larvae challenged by 100,000 bacteria of the genus Salmonella thyphimurium (strain 12023G). Bacteria were injected into the weevil hemolymph, using a Nanoinject II appartus (Drummond). Larvae were then incubated 3, 6 and 9 hours at 27.5 C and 70% rh. For each replicate, 5 larvae incubated 3h were pooled with 5 larvae incubated 6h and 5 larvae incubated 9h, then stored at -80 C until RNA preparation. 2. RNA isolation and libraries construction for RNAseq Total RNA from the 15 whole larvae was extracted with Trizol reagent (Invitrogen) following the manufacturer s instructions. RNA was incubated with 1 U/ g of RQ1 RNase-free DNase for 30 min, at 37 C then purified with RNeasy micro kit (QIAGEN) clean-up protocol. Two independent replicates of 1 g of RNA sample were provided to ProfileXpert (Lyon, France) and sequenced on an Illumina HiSeq2000 instrument following the manufacturer s protocol and generate paired-end data (51 bp reads).
