Anopheles aquasalis transcriptome in response to Plasmodium vivax invasion of midgut Overall design: A. aquasalis mosquitoes were: i) membrane-fed on P. vivax infected blood, ii) membrane-fed on blood with inactivated gametocytes or iii) fed on 10 % sugar solution. 18-24 hours after feeding, mosquitoes were dissected and pools of 30 midguts were collected from each group. Three independent biological replicates of each experiment were performed.

Construction protocol: Mosquito midguts were collected from mosquitoes submerged in ice-cold PBS and transferred to RNAlater (Ambion). RNA was harvested using Trizol reagent and treated with RNase-Free DNase I (Qiagen). RNA integrity was confirmed using 2100 Bioanalyzer (Agilent) and total RNA was used for library preparation and sequencing. RNA libraries were prepared for sequencing using standard Illumina protocols
