Illumina HiSeq technology was used to generate mRNA profiles from Pisolithus microcarpus Si9, MW3, R4 or R10 ectomycorrhizal root tips compared to their respective free-living mycelium. Overall design: Ectomycorrhizal root tips and control mycelium were harvested after 4 weeks of contact and used for RNA extraction.

Construction protocol: RNA was extracted using the Qiagen RNeasy Plant Kit using the RLC lysis buffer supplemented with 25 mg/mL PEG 8000 and following manufactuers instructions thereafter. RNA quantity was analyzed using the QuBit broad spectrum RNA kit as per manufacturers instructions and RNA quality was verified using the BioAnalyzer platform (Agilent Technologies Inc.). cDNA libraries were prepared for sequencing using standard Illumina protocols by the University of Western Sydney Next Generation Sequencing Facility (Richmont,Australia)
