SRX6438777: RNA-sequencing of Drosophila yakuba adult male brain

Design: Prior to the dissections, all fly lines were expanded and maintained at 24C under an undisturbed 12:12 light/dark cycle in low density vials with fly food. All individuals were mated and precisely at 4-days-old. Flies were very briefly anesthetized with CO2 and heads were collected with a clean razor blade. Heads were transferred to a glass dissection plate with PBS for brain collection. Dissections were performed within a 3-hour window always at the same circadian zeitgeber time (ZT1-ZT4; in this experiment the light was turned on at 8 am, and the dissections were performed between 9 am and 12 pm within a three-hour interval). Male and female brains of the same line were dissected in parallel the same day at the same time. Dissected brains were immediately transferred into a low retention Eppendorf tube containing 100 mL Trizol (Invitrogen) and kept frozen at -20C until RNA extraction. We collected 3 biological replicates for each genotype by sex combination, and each replicate comprised of a pool of exactly 15 brains.
