SRX11794064: Targeted Sequence Capture from Taeniatherum caput-medusae total leaf DNA
1 ILLUMINA (NextSeq 500) run: 16.3M spots, 4.7G bases, 1.8Gb downloads

Design: Genomic DNA was sheared to ~400 basepairs and used to build Illumina-compatible sequencing libraries. Targeted sequence capture was used to enrich coding regions from amplified libraries using exon-specific baits designed from published barley and wheat genomes. Post-capture libraries were amplified and paired-end sequenced on an Illumina NextSeq 500.

The present analysis builds upon past molecular analyses of the wheat tribe through analyses of exon sequence data from over 1000 nuclear loci obtained through targeted sequence capture, along with a matched sample of nearly-complete chloroplast genomes. The results highlight (1) strongly-supported cpDNA-based relationships; (2) several well-supported "nuclear genome" relationships among major genera; (3) multiple lineages that remain difficult to place with confidence; (4) similarities and differences among nuclear data subsets based on chromosome arms; and (5) similarities and broad differences between the chloroplast and combined-nuclear trees.
