Design: 1) Enrichment of eukaryotic mRNA with magnetic beads with Oligo (dT);(2) Random interruption of mRNA by addition of Fragmentation Buffer;(3) Synthesis of the first cDNA strand and the second strand using mRNA as a template and cDNA purification;(4) The purified double-stranded cDNA is then end-repaired, A-tailed and ligated to sequencing junctions, followed by fragment size selection using AMPure XP beads;(5) Final enrichment by PCR to obtain cDNA libraries
