Design: RNA was isolated using TRIzol (Thermo-Fisher Scientific). Library construction was performed using the TruSeq Stranded Total RNA with Ribo-Zero Human/Mouse/Rat kit (Illumina). The MiSeq Reagent Kit v3 (Illumina) was used for paired-end sequencing on a HiSeq 2000 platform (Illumina).

Study: RNA-seq of MDA-MB-231 cells upon knockdown of ASncmtRNA

Knockdown of the antisense noncoding mitochondrial RNAs (ASncmtRNAs) triggers proliferative arrest and apoptotic death of tumor cells from different tissue origins, without affecting normal cells. We aimed to study transcriptomic changes induced by ASncmtRNA knockdown in order to study the mechanisms underlying the effects on tumor cells, specifically in MDA-MB-231 breast cancer cells. MDA-MB-231 cultures were transfected with an antisense oligonucleotide (ASO-1537) targeting the antisense noncoding mitochondrial RNAs (ASncmtRNAs) or with a control ASO (ASO-C) or left untreated, for 24 h. Total RNA was collected and subjected to paired-end Illumina RNA sequencing. Experiments were performed in triplicate.
