Design: Samples were taken from two heights (1.4m and 3.8m from the base) in the reactor and split into two fractions each. Of the resulting four fractions, the granular biomass of two fractions was washed whereas the other two fractions were untreated. Of each of the four samples, DNA was extracted using two methods (organic solvent extraction & mobio powersoil kit) resulting in 8 samples for sequencing. Library preparation was performed using the kits and protocols supplied by life technologies, using enzymatic shearing and E-gel size selection. The organic solvent samples were sequenced twice, resulting in 12 runs
