Array description: The custom microarrays are manufactured by the Laboratory of Human Bacterial Pathogenesis (LHBP), Rocky Mountain Laboratories (RML), of the National Institute of Allergy and Infectious Diseases, National Institutes of Health (NIAID/NIH).
Microarrays are printed with a Bio-Rad VersArray ChipWriter Pro robotic arrayer that is fitted with a 48 pin printhead and 32 SMP3 print tips from Telechem (http://www.arrayit.com). The GAS microarray probe set is based on ~500 bp PCR products for each open reading frame (ORF) from GAS strain SF370 (AE004092) and unique ORFs from GAS strains MGAS 8232 (AE009949), and MGAS 315 (AE014074). The probes are distributed into 384-well polypropylene plates (Whatman polyfiltronics) and dissolved in 3X SSC print buffer at a normalized 25 micromolar concentration. The microarray spot densities are 200 micrometers (center-to-center rows) and 240 micrometers (center-to-center columns). The microarrays are printed on Corning GAPS coated slides and post-processed according to the procedures recommended by the manufacturer.
