Patrick Wilson's laboratory, University of Chicago using ArrayIt technology (SpotBot3 microarrayer)
Manufacture protocol
1. 6-8µl of each human monoclonal antibodies at 500µg/ml are deposited in a 384 wells plate (ArrayIt) 2. the same volume of 2X Printing Buffer (ArrayIt) is added in each well. Mix well. 3. antibodies are then printed, in triplicate, on SuperEpoxy treated glass slides (ArrayIt). The same set of antibody is spotted 24 times on the same slide allowing 24 samples to be applied. 4. After the run, slides are kept at 4 degres before use.
Description
Human monoclonal antibodies spotted. The mAbs have been isolated from different individuals. Some antibodies are from the same individual (i.e., the ones starting by the same 3 numbers, for example 011) but they have been cloned from different cells (we isolate single B cells by flow cytometry and clone the heavy and light chain to make the mAbs).
