Purified recombinant proteins were diluted at final concentration of 0.5 mg/ml in 40% glycerol. Recombinant E. coli GMMA and OMV from NZ98/254 were prepared at 1 mg/ml in 20% glycerol and then serially diluted in 20% glycerol (step-2 dilutions). Samples were transferred into 384-well plates. Two droplets of 0.1 nL protein solution was deposited per spot on nitrocellulose-coated slides (FAST slides, Maine Manufacturing) using the non-contact printer Marathon Argus Spotter (Arrayjet). Printing was performed in a cabinet with controlled humidity and temperature (50-55% and 18°C, respectively). Each protein was printed in replicates on each slide. Printed protein microarrays were kept at room temperature for 5 hours, dried under a stream of nitrogen and store at 4°C. Only the highest concentration of the vesicles was included in the final dataset.
