Array elements were generated for the annotated S. pombe genome using PCR and specific primer pairs. Primers, chosen to ensure unique amplification, varied in length, melting temperatures, and GC contents. Amplicons, containing exclusively exon sequences, were verified against the genome to avoid significant similarity with other sequences. Additionally, various genomic features including mitochondrial genes, pseudogenes, and RNA genes, totaling 5269 elements, were also amplified. Control elements from B. subtilis were included. The PCR process involved two rounds, utilizing gene-specific primers and a universal sequence, with products verified for consistency. Post-PCR, the products underwent filtration, arraying, and storage, with duplicate spots printed per slide for robust measurements. Quality control was conducted through hybridization tests.
