Oligonucleotides are tandem complements of mature miRNA sequences and snoRNA sequences. they contain 5' C6 amino linkers to allow binding the the aldehyde microarray slide upon drying. Oligonucleotides were spotted at a 40 micromolar concentration in 2.4x SSC. After spotting, slides remained in a high-humidity chamber for 4 hours, and were then allowed to dry overnight. Dried slides were washed twice 0.2% SDS solution for 2 minutes each time, twice in H2O for 2 minutes each time, for 5 minutes in sodium borohydride solution (375 ml 1x PBS, 1.3 g sodium borohydride, 125 ml 100% ethanol after sodium borohydride has dissolved). After sodium borohydride wash, the SDS and H2O washes were repeated. Slides were spun dried and stored at 4 degrees celcius in a dry environment.
